According to the United States Census Bureau, Deferiet has a total area of 0.73 square miles (1.9 km2), of which 0.66 square miles (1.7 km2) are land and 0.1 square miles (0.2 km2), or 11.21%, are water.

Deferiet is located on New York State Route 3, which leads west 12 miles (19 km) to Watertown, the Jefferson county seat, and southeast 6 miles (10 km) to Carthage. Route 3A intersects NY-3 in Deferiet and leads east 5 miles (8 km) to a second junction with NY-3 in the hamlet of Fargo.
